Oncogenicity Studies and Drug Screening

The Ba/F3 cell line is mouse pro-B cell requiring interleukin-3 (mIL3) for survival and proliferation.  Ba/F3 cells may grow without mIL3 while the Ba/F3 cell line transduced and transformed by an oncogene.  Therefore, the Ba/F3 cell line has been a popular system to evaluate oncogenicity of genes and mutations, and efficacies of targeted drugs.

Biosettia have established a workflow including cloning the genes of interest and mutants, generating lentiviruses and stable lines, mIL3 withdrawal to confirm the oncogenicity and drug screening.

Flowchart illustrating genetic engineering process, including lentiviral vector production, viral transduction into cells, integration and gene expression, cell proliferation, and testing with a drug dispenser, plate reader, and cell titer assay.
